Dyspeptic disorders against the background of delayed gastric emptying, gastroesophageal reflux disease: feeling of overflow in the epigastrium, bloating, pain in the upper abdomen, belching, flatulence, heartburn with or without gastric contents throwing up into the mouth. Nausea and vomiting of various genesis (including against the background of functional and organic diseases, infections, toxemia, radiation therapy, dietary disorders, drug genesis, including. intake of dopamine-levodopa agonists, bromocriptine, endoscopic and X-ray gastrointestinal examinations); hiccups, gastrointestinal atony (including postoperative); the need to accelerate peristalsis during X-ray gastrointestinal examinations.

Contraindications

Hypersensitivity, gastrointestinal bleeding, mechanical intestinal obstruction, gastric or intestinal perforation (stimulation of peristalsis may aggravate these conditions), hyperprolactinemia, prolactinoma; children under 5 years of age and children under 20 kg body weight – for tablets.

Limitations to use

Renal and/or hepatic failure, pregnancy, breast-feeding. Use during pregnancy and lactation. It is possible if the expected effect of the therapy exceeds the potential risk for the fetus and the child (studies in pregnant women have not been conducted, low concentrations of domperidone are found in the breast milk of women).

Side effects of the substance Domperidone

  • Gastrointestinal organs: spasm of smooth muscles of the gastrointestinal tract, dry mouth, stomatitis, thirst, heartburn, change of appetite, constipation/diarrhea.
  • Nervous system and sense organs: extrapyramidal disorders (in children and patients with increased permeability of the GEB), headache, asthenia, irritability, nervousness, drowsiness, leg cramps, lethargy, conjunctivitis (itching, redness, pain, swollen eyes).
  • Allergic reactions: skin rash, itching, urticaria, swelling (face, hands, shins or feet).
  • Other: increased plasma prolactin levels, galactorrhea, gynecomastia, menstrual disorders, mastalgia, changes in urinary frequency, dysuria (burning, difficulty and pain during urination), palpitation.

Interaction

Precautions for taking emetrol (domperidone)

Antacids and antisecretory drugs reduce bioavailability, anticholinergic agents weaken the effect of domperidone. CYP3A4 isoenzyme inhibitors of cytochrome P450 (azole antifungal medicines, antibiotics of macrolide group, HIV protease inhibitors) can block metabolism of domperidone and increase its level in plasma (joint use requires caution). Use with caution concomitantly with MAO inhibitors. The effect on absorption of concomitantly used drugs with delayed release of the active substance is not excluded. Domperidone does not affect the blood levels of paracetamol and digoxin.

Overdose

Symptoms: somnolence, arrhythmia, disorientation, extrapyramidal disorders (especially in children), BP decrease.

Treatment: use of activated charcoal, in case of extrapyramidal reactions – anticholinergic agents or antihistamines with anticholinergic activity, drugs used for treatment of Parkinsonism. There is no specific antidote.

Precautions of the substance Domperidone. Should not be prescribed for prevention of postoperative vomiting.
